Comparison of Dna-extraction Methods and Selective Enrichment Broths on the Detection of Salmonella Typhimurium in Swine Feces by Polymerase Chain Reaction (pcr)

چکیده
The aim of this study was to compare different DNA-extraction methods and selective enrichment broths for their effectiveness to detect Salmonella Typhimurium in artificially inoculated swine feces samples (100 CFU/ g) by polymerase chain reaction. After enrichment in Rappaport-Vassiliadis, selenite cystine or MüllerKauffmann tetrathionate, aliquots were used for DNA extraction by three different methods: boilingcentrifugation, phenol-chloroform and salting-out. Aliquots of extracted DNA were then used as template in PCR. The selective enrichment broths had no effect on the efficiency of PCR when boiling-centrifugation and salting-out were used. On the other hand, phenol-chloroform was superior (P<0.05) when combined to Rappaport-Vassiliadis. Considering cost and efficiency parameters, we encourage the use of Müller-Kauffmann tetrathionate broth in combination with boiling-centrifugation DNA-extraction procedure.

Potential Use of DNA Aptamer-Magnetic Bead Separation-PCR Assay for Salmonella Detection in Food
Background: Salmonella is one of the most common food-borne pathogens that can cause illness. In this study, the sensitivity and the specificity of Aptamer-Magnetic bead Separation-Polymerase Chain Reaction (AMS-PCR) method were determined for Salmonella spp. detection. Methods: Different concentrations of Salmonella enterica were mixed with streptavidin-magnetic beads coated with biotinylated...

Previously designed endpoint PCR has been adapted for use with real time PCR to detect the presence of Salmonella typhimurium and Salmonella enteritidis in fish. Optimization of a standard curve in the presence of herring sperm DNA as background matrix indicated that the real time PCR highly efficient with the Pearson coefficient of determination (R) value = 0.99937 and slope (M) value = -3.44....

Introduction: In Venezuela, acute diarrheic syndrome (ADS) is a primary cause of morbimortality, often involving the Salmonella genus. Salmonella infections are associated with acute gastroenteritis, one of the most common alimentary intoxications, and caused by the consumption of contaminated water and food, especially meat. Methods: Conventional and molecular methods were used to detect Salmo...
